      <image:title>Prostate Cancer</image:title>
      <image:caption>Acquired resistance to targeted therapies in cancer is a rising unmet clinical need. In prostate cancer (PCa), the success of next-generation androgen receptor (AR) pathway inhibitors has been hampered by the development of drug resistance. This acquired resistance involves, in many cases, the reactivation of the AR axis through several different mechanisms. However, an alternative mechanism of resistance is driven by the reprogramming of prostate cancer cells to undergo lineage plasticity to adopt an AR-independent state and to acquire a neuroendocrine phenotype, that allow them to grow and survive and escape AR-therapy. More potent and sustained AR targeting has driven an increased incidence of neuroendocrine prostate cancer (NEPC), which is an extremely aggressive, highly proliferative and metastatic PCa variant. Therefore, understanding the molecular mechanisms that govern NEPC differentiation is a pressing unmet clinical need. We are interested in unveiling the signaling and metabolic pathways that account for these changes in cell identity during therapy resistance with the goal of identifying new targets that either restore sensitivity to current therapeutics or that exploit new previously unanticipated tumor cell vulnerabilities. Our recent studies have discovered that the downregulation of PKCl/i is a hallmark of human NEPC patients. We demonstrated that the loss of PKCl/i results in the upregulation of the serine and one-carbon pathway that leads to increased production of S-adenosine methionine, universal and obligated methyl donor for DNA and histone methylation. Our present work aims to unravel the epigenetic mechanisms linking metabolic reprogramming by PKCl/i to cell lineage alterations in PCa and possibly therapy resistance in other types of cancers. We are also investigating how the remodeling of the tumor microenvironment, particularly the stromal fibroblasts, facilitates therapy resistance, and how the metabolic interplay between cancer-associated fibroblasts and the epithelial compartment of the tumor informs new therapies.</image:caption>

      <image:caption>Maria T. Diaz-Meco studies the molecular and cellular mechanisms that control therapy resistance, with a particular interest in the regulation of lineage plasticity in prostate cancer. Her current approach is to identify metabolic and epigenetic vulnerabilities that will serve as therapeutic targets for new anti-cancer medicines that selectively impact tumor cell growth and survival. Diaz-Meco recently identified the metabolic pathways underlying the acquisition of therapy-resistance in patients of the most aggressive form of metastatic prostate cancer. These studies unraveled potential targets in the control of the metabolic and epigenetic programs whose therapeutic manipulation could restore sensitivity to androgen-deprivation therapy to patients that have become refractory to this type of treatment. Maria is also interested in the metabolic impact of stromal reprogramming and the crosstalk of the tumor cell and its microenvironment during tumor initiation and progression in prostate cancer. The overall goal of her projects is to study metabolism and inflammation in prostate cancer, understand this disease's biology, and identify potentially novel therapeutic targets. Maria T. Diaz-Meco earned a bachelor’s degree in Chemistry and her Ph.D. in Biochemistry &amp; Molecular Biology at the University Complutense in Madrid, Spain, with graduate training studies at the National Cancer Institute. She was an Associate Professor in the National Research Council of Spain when she was recruited in 2006 to the Genome Research Institute at the University of Cincinnati. In 2010, she was appointed Professor of Cancer and Cell Biology at the University of Cincinnati Medical College, and in 2011 she joined the Sanford Burnham Prebys Medical Discovery Institute as Professor in the Cancer Metabolism and Signaling Networks Program. In 2020, she was recruited to Weill Cornell Medicine as Professor in the Department of Pathology and Laboratory Medicine. Since January 2021, she is a Homer T. Hirst III Professor of Oncology in Pathology at WCM.</image:caption>

      <image:caption>Jorge Moscat is a Cancer Biologist with expertise in the cellular and molecular mechanisms that control the initiation and progression of tumors. He focuses on liver and colorectal cancer and has made impactful contributions to understanding the metabolic and immunological landscape of these tumors and their response to immunotherapy. His research aims to identify non-oncogenic vulnerabilities in cancer, with a particular interest in the interface between inflammation and metabolism. Jorge’s work led to the identification and characterization of several novel signaling molecules such as the autophagy and PB1-containing signaling adaptors p62/SQSTM1 and NBR1, and the PB1 kinases PKCz and PKCl/i.  He has served in numerous scientific and leadership positions, including being an elected member of the prestigious European Molecular Biology Organization. Jorge Moscat presently is a Homer T. Hirst III Professor of Oncology in Pathology and Vice-Chair for Experimental Pathology at the Weill Cornell Medical College. Before joining Cornell, Jorge has served in several leadership positions at the Sanford Burnham Prebys Medical Discovery Institute in La Jolla (California), including Deputy Director of the Cancer Center and also as the Institute’s Director of Metabolism Initiatives. Before that, Jorge was a Professor and Chairman of the Department of Cancer and Cell Biology and Associate Director of the Cincinnati Cancer Consortium at the University of Cincinnati Medical College. He also served in several positions in the National Research Council of Spain, in which he was a Professor and Director of the Institute for Molecular Biology.</image:caption>

      <image:title>Metabolism and Cancer</image:title>
      <image:caption>Tumor cells have an extremely elevated need for nutrients to sustain their high anabolic requirements necessary to maintain their enhanced proliferative rate. They are tremendously plastic and can reprogram their metabolism to more effectively use nutrients, when they are abundant, and reframe several key metabolic pathways to resist nutrient deprivation when they become aggressive and in poorly vascularized microenvironments. The study of these mechanisms opened the possibility of identifying metabolic dependencies and addictions in each type of cancer cell, which in fact become vulnerabilities that can be targeted therapeutically. Our laboratories actively explore these pathways and have very significantly contributed to the field by identifying several key mechanisms in the study of tumor metabolism. These included the serine biosynthetic and one-carbon pathways that are a central node in the control of cell energetics, oxidative detoxification, and epigenetic control through the generation of methyl donor groups for DNA and histone methylation. Although part of the lab focuses on the metabolism of tumor cells, there is another aspect of this question that is of paramount importance and that deals with the question of total body metabolism, the metabolic syndrome, and its relationship with cancer. Our labs were the first to report the role of p62/SQSTM1 in adipocyte function. We showed that the loss of p62 in adipocytes triggered increased adiposity and obesity in mice. Interestingly, this has very important effects on cancer. Thus, we demonstrated that the loss of p62 in adipocytes results in increased osteopontin secretion, which mediates tumor fatty acid oxidation and invasion, leading to aggressive metastatic prostate cancer. Importantly, while promoting tumor malignancy, the adipocyte’s p62 deficiency promotes a general shutdown of energy-utilizing pathways in adipocytes through mTORC1 inhibition, which supports nutrient availability for cancer cells. These results exemplified the symbiotic collaboration between adipocytes and tumor cells in which p62 emerges as a central regulator.</image:caption>
